Elimination of inhibitors of alkaline phosphatase from 2-amino-2-methyl-1-propanol.

چکیده
When 2-amino-2-methyl-1-propanol buffer solutions are extracted with chloroform, impurities inhibiting alkaline phosphatase are selectively removed and this source of variation is thereby greatly decreased. We also studied the effect of adding zinc sulfate to this buffer. A combination of chloroform extraction and zinc sulfate, added to give concentrations from 5 to 10 mumol/L, results in buffer of high and consistent quality. The alkaline phosphatase activities of 12 reference sera of different origin were determined with use of the purified buffers. Those sera, which closely resembled human serum in terms of alkaline phosphatase isoenzyme composition, behaved similarly to human serum. Use of such control sera is strongly recommended.

Effect of incubation with Mg2+ on the measurement of alkaline phosphatase activity.
Human serum incubated with 2-amino-2-methyl-1-propanol (860 mmol/liter) and magnesium ion (270 mumol/liter) at pH 10.35 showed greater alkaline phosphatase (EC 3.1.3.1) activity than if an equal amount of magnesium ion was added at the time of measurement. The apparent increase is due in part to a slight lability of serum alkaline phosphatase in 2-amino-2-methyl-1-propanol, which is prevented b...
متن کاملAn impurity in the buffer 2-amino-2-methyl-1-propanol, which correlates with depression of measured alkaline phosphatase activity.
Recent findings indicate that the buffer 2-amino-2-methyl-1-propanol, widely used in clinical measurement of alkaline phosphatase activity, contains an impurity that inhibits the activity of the enzyme. Analysis of several lots of 2-amino-2-methyl-1-propanol has demonstrated the presence of a material, the concentration of which correlates well with alkaline phosphatase activity. Mass spectral ...
متن کاملMeasurement of alkaline phosphatase activity: characterization and identification of an inactivator in 2-amino-2-methyl-1-propanol.
An inactivator of alkaline phosphatase (EC 3.1.3.1) in 2-amino-2-methyl-1-propanol is demonstrated and characterized. This time-dependent inactivation results from chelation of enzyme-bound Zn2+; it is reversed by addition of Zn2+ and, to a lesser extent, other divalent metal ions. Cu2+ is an effective spectral indicator and can be used to determine the presence and quantity of inactivator. Dat...
متن کاملAn automated p-nitrophenylphosphate serum alkaline phosphatase procedure for the "Robot Chemist".
A new, precise, and accurate procedure is described for the automated determination of serum alkaline phosphatase on the Robot Chemist. The substrate, p-nitrophenylphosphate in 2-amino-2-methyl-1-propanol, offers the prime advantage of providing directly its own chromogen, p-nitrophenol, following enzyme action. Other advantages are the small sample volumes required and linear reaction rates. C...
متن کاملQuantitative method for determining serum alkaline phosphatase isoenzyme activity: estimation of intestinal component.
Intestinal alkaline phosphatase activity was measured using levamisole inhibition, and results were compared with a previously reported method using L-phenylalanine. Sixty two per cent intestinal, 39% placental, and 1.3% of either bone or liver alkaline phosphatase activity remained when alkaline phosphatase activity was inhibited in a 2-amino-2-methyl-1-propanol (AMP) buffer reagent system wit...
